reducing agent:
Due to the presence of inter-protein disulfide bonds, reducing agents are generally used at the time of solubilization. Reducing agents are generally used in concentrations of 50-100 mM 2-BME or DTT, as are 5 mM concentrations. Under more extensive conditions, a concentration of 5 ml / l can be used. The concentration of reducing agent has nothing to do with the number of protein disulfide bonds, while some of the proteins without disulfide bond have no effect on reducing agents such as the solubilization of bovine growth hormone inclusion bodies. For the solubilization of certain inclusion bodies without the disulfide bond of the target protein, the use of reducing agents is sometimes necessary, possibly due to the inclusion body disulfide-solubilizing inclusions.
Refolding:
By slowly removing the denaturant, the target protein is restored from its denatured fully extended state to the normal folded structure, while the reducing agent is removed to allow the disulfide bond to form normally. Generally, the refolding process begins when the urea concentration is about 4M, and ends at about 2M. For guanidine hydrochloride, we can start from 4M, to 1.5M renaturation process has ended.
Methods often used in renaturation are:
Dilution refolding: directly add water or buffer, placed overnight, the disadvantage is a larger volume increase, denaturant dilution rate too fast, not easy to control.
Dialysis refolding: the advantage is not to increase the volume, by gradually reducing the concentration of liquid to control the rate of denaturant removal, it was easy to form a precipitate, and is not suitable for large-scale operation, can not be applied to the scale of production.
Ultrafiltration renaturation: more use in production, larger, easy to control the speed of dialysis, the disadvantage is not suitable for the case of small sample size, and some proteins may be irreversible denaturation in the ultrafiltration process.
Refolding: This is a more recent and successful method of refolding used in production. For example, North China Pharmaceutical's refolding of G-CSF is said to be performed by refolding on a column. Commonly used in the renaturation of the chromatographic methods SEC, HIC and so on.
