日本理化學(xué)研究所與日本東北大學(xué)的研究小組對開角型青光眼實(shí)施了全基因組關(guān)聯(lián)分析（GWAS），認(rèn)定了7個(gè)新的易感基因區(qū)域。

青光眼是視神經(jīng)受損的眼病，在日本占失明原因的*位。其主要類型為開角型青光眼，目前對該病的遺傳因素尚知曉不多。

研究小組發(fā)表在英國《人類分子遺傳學(xué)》雜志上的論文稱，他們利用日本生物銀行收集的3980名開角型青光眼患者和18815名對照群組為對象，對全體人類基因組分布的約600萬個(gè)單核苷酸多態(tài)性（SNP）進(jìn)行了全基因組關(guān)聯(lián)分析。此外，對已認(rèn)定與開角型青光眼強(qiáng)相關(guān)的SNP，通過兩個(gè)獨(dú)立的群組（患者3398名、對照組17570名）進(jìn)行了再現(xiàn)性印證。結(jié)果發(fā)現(xiàn)，7個(gè)新的基因區(qū)域?qū)η喙庋郯l(fā)病有影響。小組用這些基因區(qū)域與其他人種（患者8357名、對照組38100名）的發(fā)病風(fēng)險(xiǎn)進(jìn)行驗(yàn)證，認(rèn)定其中2個(gè)SNP參與亞洲系人種、4個(gè)SNP參與歐洲系人種發(fā)病。對相關(guān)基因特征進(jìn)行路徑分析，查明了上皮生長因子受體信號相關(guān)基因群影響青光眼發(fā)病。

研究小組還對過去報(bào)告的開角型青光眼相關(guān)的7種特征進(jìn)行了遺傳學(xué)評價(jià)，發(fā)現(xiàn)開角型青光眼與Ⅱ型糖尿病和心肌梗塞、缺血性中風(fēng)等心血管疾病有共同的遺傳背景，這意味著有些人生來易患此病。他們將進(jìn)一步研究全身狀態(tài)和青光眼的關(guān)系，期望為預(yù)防醫(yī)學(xué)作出貢獻(xiàn)。

The research team of Japanese physicochemical Institute and Tohoku University Japan carried out genome-wide association analysis (GWAS) for open angle glaucoma, and identified 7 new susceptible gene regions.

Glaucoma is the eye disease of the optic nerve, which is the first cause of blindness in Japan. Open angle glaucoma is the main type of glaucoma, and the genetic factors of the disease are not known much at present.

The research team published in the British Journal human molecular genetics, "" the paper said, they use the Japanese bank collected 3980 biological open-angle glaucoma patients and 18815 control group as the object, the whole human genome distribution of about 6 million single nucleotide polymorphisms (SNP) conducted a genome-wide association study. In addition, SNP, which was strongly associated with open angle glaucoma, was reproduced by two independent groups (3398 patients and 17570 controls). The results showed that 7 new gene regions had an impact on the incidence of glaucoma. The team used these gene regions to verify the risk of other races (8357 patients and 38100 controls), and identified 2 SNP participating in Asian race and 4 SNP participating in European race. A path analysis of the related gene features was carried out to determine the influence of the epithelial growth factor receptor signal related gene group on the incidence of glaucoma.
